In our brain, every memory and decision begins with a neuron communicating a signal through a synapse. Numerous neurological diseases have been associated with synaptic dysfunction, thus understanding how synaptic proteins are regulated is of critical importance to human health. While the physical composition and architecture of synapses have been extensively explored, the mechanisms that control synaptic gene expression are unclear. We have recently discovered a novel transcriptional program, controlled by neuronal activity, which drives synaptogenesis. This work was among the first to demonstrate a link between activity and genetic programs to sculpt synapse formation in vivo.

In our lab, we leverage molecular biology, cell biology, proteomics, genomics, and genetics to discover fundamental mechanisms that control synaptic gene expression at the resolution of a single neuron. By identifying these mechanisms and investigating how they are controlled throughout aging, we will gain valuable knowledge that could be applied to improve outcomes of individuals with neurological disorders with altered synaptic protein levels.
